Job Title: Laboratory Technician
Location: NAC Center at TSRI, La Jolla, California

Position Summary:

• The candidate will perform state-of-the-art molecular biology and recombinant protein expression, production, purification and characterization of HIV-1 envelope glycoproteins (Env) or related proteins.
• The HIV-1 Env will be used both as immunogens and as reagents for analyzing anti-Env immune responses elicited in relevant small animal models, non-human primates and humans.
• The candidate may also study the mechanisms of neutralization by different monoclonal antibodies and may work with HIV-1 pseudoviruses under BSL3 laboratory safety conditions.
• The candidate will be able to design, execute experiments and interpret the data in an independent manner or with limited supervision.
• Minimum qualifications include a M.S. or B.S. degree in molecular biology or virology or related areas of biological sciences.
• The candidate will preferably possess 3 years of relevant laboratory research experience, but other highly qualified candidates will be considered.


Key Responsibilities:
• Standard mutagenesis and cloning to generate protein expression vectors
• Expression and purification of HIV-1 Env and other proteins on a routine basis
• Perform FACS-based binding assays
• Perform ELISAs and immunoprecipitation assays
• Perform viral neutralization assays
• Maintain cells lines in culture for transfection, virus production and as neutralization targets
• Prepare pseudotyped virus stocks under BSL3 conditions
• Isolate PBMCs from selected blood samples
• Organize effectively and discuss data
• Order reagents and supplies and display good laboratory practices



Experience and Skills:
• Molecular biology: RNA/DNA purification, PCR, site-directed mutagenesis…
• Biochemistry: protein purification, FPLC, immunoprecipitation, SDS-Page, Western Blots…
• Cellular biology and immunology: cell culture, cell purification, ELISAs, FACS
• Experience with gel filtration chromatography is a plus
